Chickens were immunized with a synthetic peptide containing the human c-myc epitope tag – EQKLISEEDL – conjugated to keyhole limpet hemocyanin (KLH).The sequence of this peptide corresponds to residues #425-434 of the human c-myc proto-oncogene protein (NCBI NP_002458.2).After repeated injections, immune eggs were collected from the hens, and the IgY fractions were purified from the yolks.These IgY fractions were then affinity-purified using an agarose matrix to which the cognate peptide was attached.Eluates of this column were dialyzed against PBS, and then conjugated to fluorescein using its isothiocyanate form (FITC).Fluorescein-labeled antibody was separated from free FITC using gel exclusion chromatography (Sephadex G-50), and the protein concentration was adjusted to 0.1 mg/mL in PBS with 0.02% sodium azide.Finally, the preparation was filter-sterilized.

Store at 4°C in the dark. Under these conditions, the antibodies should have a shelf life of at least 12 months (provided they remain sterile). For longer storage periods, dilute the antibodies with sterile glycerol and store at -20°C. Do not freeze these antibodies unless you want to store them for longer periods of time.
